2'-Deoxyguanosine compounds represented by formula (2), may be prepared by desulfurizing an 8,2'-anhydro-8-mercapto-9-.beta.-arabinofuranosylguanine compound represented by formula (1) in a solvent in the presence of a base using nickel or a nickel alloy. wherein R.sup.1, R.sup.2, R.sup.4, and R.sup.5 are each independently a hydrogen atom or a hydroxyl-protecting group, and R.sup.3 and R.sup.6 are hydrogen atoms or amino-protecting groups.
