Methods for enhancing thrombolysis in a mammal

   
   

It has been found that agents which inhibit the interaction between the band 3 protein and its ligand (hereafter sometimes referred to as "interaction inhibitors" or "inhibitors"), CD36/thrombospondin, can also be used to enhance thrombolysis. The inhibitors can be peptides which contain sequences present in exofacial loops of the band 3 protein, or can be non-natural, D-isomer forms of the same sequences, or can be peptides, peptidomimetics, or non-peptidic molecules which interfere with band 3 protein--ligand interactions. One preferred group of such inhibitors comprises peptides characterized by the sequence motif Z.sup.2 Z.sup.3 Z.sup.2 UX.sup.- UUUX.sup.- (SEQ ID NO:44), wherein Z.sup.2 represents a hydrophobic residue, U represents unobstructive residues, Z.sup.3 is either Z.sup.2 or an unobstructive residue and X.sup.- represents negatively charged residues. Another preferred group of such inhibitors comprises peptides characterized by the sequence motif Z.sup.1 UKUUUX.sup.+ (SEQ ID NO:45), wherein Z.sup.1 is selected from the group consisting of tyrosine, phenylalanine and alanine; K is a lysine residue; U represents unobstructive residues; and X.sup.+ is a positively charged residue. The reverse sequences of these peptides are also active. The inhibitors of the subject method can be administered alone or in combination with one or more additional therapeutic agents. Administration can be oral, intravenous or by other means, in amounts between about 0.1 mg/kg to about 20 mg/kg.

Ein bevorzugte Gruppe solcher Hemmnisse enthält die Peptide, die durch das Reihenfolge Motiv Z.sup.2 Z.sup.3 Z.sup.2 UX.sup. gekennzeichnet wurden - UUUX.sup. - (FOLGENDE Identifikation NO:44), worin Z.sup.2 einen hydrophoben Überrest darstellt, stellt U unobstructive Überreste dar, ist Z.sup.3 entweder Z.sup.2 oder ein unobstructive Überrest und ein X.sup. - stellt negativ belastete Überreste dar. Eine andere bevorzugte Gruppe solcher Hemmnisse enthält die Peptide, die durch das Reihenfolge Motiv Z.sup.1 UKUUUX.sup.+ gekennzeichnet werden (FOLGENDE Identifikation NO:45), worin Z.sup.1 von der Gruppe vorgewählt wird, die aus Tyrosin, Phenylalanin und Alanin besteht; K ist ein Lysinüberrest; U stellt unobstructive Überreste dar; und X.sup.+ ist ein positiv belasteter Überrest. Die Rückreihenfolgen dieser Peptide sind auch aktiv. Die Hemmnisse der vorbehaltlichen Methode können einem oder mehr zusätzlichen therapeutischen Mitteln alleine oder im Verbindung mit ausgeübt werden. Leitung kann mündlich, Intravenous oder durch andere Mittel, in den Mengen zwischen ungefähr 0.1 mg/kg zu ungefähr 20 mg/kg sein.
