Human .beta.-globin mutant polypeptides comprise the amino acid sequence of
normal human .beta.-globin modified by (i) the substitution or deletion of
Cys at amino acid positions 93 and 112, and (ii) the substitution of a Cys
fore a non-Cys amino acid at one other position in the polypeptide,
preferable Ser at amino acid position 9. Modified human hemoglobins
containing this or other .beta.-globin mutations, and/or an .alpha.-globin
mutation, are provided, as well as polymers of such modified hemoglobins.
Nucleic acids, vectors, and transformed host cells for producing the
.beta.-globin mutants are provided.
Menschliche beta.-globin Durch Mutation entstehende Variation Polypeptide enthalten die Aminosäurereihenfolge vom normalen menschlichen beta.-globin, das durch (i) den Ersatz oder die Auslassung von Cys in Aminosäurepositionen 93 und 112 geändert wird, und (ii) der Ersatz eines Cys Vorderteils eine non-Cys Aminosäure in einer anderen Position im Polypeptid, vorzuziehendes Ser in Aminosäureposition 9. Die geänderten menschlichen hemoglobins, die dieses oder andere beta.-globin Veränderungen enthalten, und/oder eine alpha.-globin Veränderung, werden, sowie Polymer-Plastiken solcher geänderter hemoglobins zur Verfügung gestellt. Nukleinsäuren, Vektoren und umgewandelte Wirtszellen für das Produzieren der beta.-globin Durch Mutation entstehende Variationen werden zur Verfügung gestellt.
