The present invention relates to the field of the Medicine, in particular
with the chemical synthesis of oligosaccharide mixtures derived of
ribose-ribitol-phosphate, which are used as active principle in vaccines
for the prevention of infections caused by Haemophilus influenzae type b
(Hib), as well as with the vaccines containing said oligosaccharide
mixtures.
The oligasaccharide mixtures obtained by chemical synthesis of the present
invention, comprise repeating units of formulae
(phosphate-ribosa-ribitol)n or (ribose-ribitol-phosphate)n of at least 5
compounds of structure A or B, which represent the repeating unit of the
capsular polysaccharide of Haemophilus influenzae type b and differ only
by n, being n a value contained between 4 and 25 (n.gtoreq.4 y.ltoreq.25),
and wherein R.sub.1 or R.sub.2 is a spacer for conjugation to a carrier,
with the condition of R.sub.1 =spacer if R.sub.2 =H, or R.sub.2 =spacer if
R.sub.1 =H.

The invention also is related with the immunogens containing such
oligosaccharide mixtures, with the vaccines containing said immunogens and
with the methods to prepare these oligosaccharides as mixtures.
Furthermore, the invention includes the use of the vaccines, alone or
combined with other vaccines, for the prevention of the infections caused
by Haemophilus influenzae type b.
A invenção atual relaciona-se ao campo da medicina, no detalhe com a síntese química das misturas do oligosaccharide derivadas do ribose-ribitol-ribose-ribitol-phosphate, que são usadas como o princípio ativo nas vacinas para a prevenção das infecções causadas pelo tipo b dos influenzae de Haemophilus (Hib), as.well.as com as vacinas que contêm misturas ditas do oligosaccharide. As misturas do oligasaccharide obtidas pela síntese química da invenção atual, compreendem repetir unidades das fórmulas (phosphate-ribosa-ribitol)n-ribosa-ribitol)n ou (o ribose-ribitol-phosphate)n-ribitol-phosphate)n ao menos de 5 compostos da estrutura A ou B, que representam a unidade repetindo do polysaccharide capsular do tipo b dos influenzae de Haemophilus e diferem somente por n, sendo n um o valor conteve entre 4 e 25 (n.gtoreq.4 y.ltoreq.25), e wherein R.sub.1 ou R.sub.2 são um espaçador para o conjugation a um portador, com a condição de R.sub.1 = espaçador se R.sub.2 = H, ou R.sub.2 = espaçador se R.sub.1 = H. ## do ## STR1 A invenção é relacionada também com os immunogens que contêm tais misturas do oligosaccharide, com as vacinas que contêm immunogens ditos e com os métodos para preparar estes oligosaccharides como misturas. Além disso, a invenção inclui o uso das vacinas, sozinho ou combinado com outras vacinas, porque a prevenção das infecções causadas por influenzae de Haemophilus datilografa b.
