Inhibition of angiogenesis by high molecular weight kininogen domain 3 peptide analogs

   
   

Peptide analogs the high molecular weight kininogen domain 3 are potent inhibitors of angiogenesis. The peptides have the formula (a) X.sub.1 -Asn-Asn-Ala-Thr-Phe-Tyr-Phe-Lys-X.sub.2, (b) X.sub.3 -Cys-Val-Gly-Cys-X.sub.4, (c) X.sub.5 -Leu-Asp-X.sub.7 -Asn-Ala-Glu-Val-Tyr-X.sub.6, or (d) Tyr-Phe-Ile-Asp-Phe-Val-Ala-Arg-Glu-Thr-Thr-X.sub.7 -Ser-Lys-Glu-Ser wherein: X.sub.1, X.sub.2, X.sub.3, X.sub.4, X.sub.5, and X.sub.6 are from zero to twelve amino acids, independently the same or different, more preferably from zero to six amino acids, and; X.sub.7 is Ala or Cys. The peptides may also comprise biologically active fragments of high molecular weight kininogen domain 3. Methods of inhibiting endothelial cell proliferation and angiogenesis are provided.

Os peptides têm a fórmula (a) X.sub.1 - Asn-Asn-Ala-Thr-Phe-Tyr-Phe-Lys-X.sub.2, (b) X.sub.3 - Cys-Val-Gly-Cys-X.sub.4, (c) X.sub.5 - Leu-Asp-X.sub.7 - Asn-Ala-Glu-Val-Tyr-X.sub.6, ou (d) Tyr-Phe-Ile-Asp-Phe-Val-Ala-Arg-Glu-Thr-Thr-X.sub.7 - Ser-Lys-Glu-Ser wherein: X.sub.1, X.sub.2, X.sub.3, X.sub.4, X.sub.5, e X.sub.6 são zero a doze aminos-ácido, independentemente os mesmos ou diferente, mais preferivelmente zero a seis aminos-ácido, e; X.sub.7 é ala ou Cys. Os peptides podem também compreender fragmentos biològica ativos do domínio elevado 3 do kininogen do peso molecular. Os métodos de inibir o proliferation e o angiogenesis endothelial da pilha são fornecidos.
