A composition for delivery of a therapeutic agent is provided. The
composition comprises: (a) a biocompatible block copolymer comprising one
or more elastomeric blocks and one or more thermoplastic blocks and (b) a
therapeutic agent, wherein the block copolymer is loaded with the
therapeutic agent. The block copolymer is preferably of the formula
X--(AB).sub.n, where A is an elastomeric block, B is a thermoplastic
block, n is a positive whole number and X is a seed molecule. The
elastomeric blocks are preferably polyolefin blocks, and the thermoplastic
blocks are preferably selected from vinyl aromatic blocks and methacrylate
blocks. According to another aspect of the invention, a medical device is
provided, at least a portion of which is insertable or implantable into
the body of a patient. The medical device comprises (a) the above
biocompatible block copolymer and (b) a therapeutic agent, wherein the
block copolymer is loaded with the therapeutic agent. According to another
aspect of the present invention, a method of treatment is provided in
which the above device is implanted or inserted into a patient, resulting
in the release of therapeutic agent in the patient over an extended
period. According to yet another aspect of the invention, a coated medical
device is provided which comprises: (a) an intravascular or intervascular
medical device and (b) a coating over at least a portion of the
intravascular or intervascular a medical device, wherein the coating
comprises the above biocompatible block copolymer.
Een samenstelling voor levering van een therapeutische agent wordt verstrekt. De samenstelling bestaat uit: (a) een biocompatibel blokcopolymeer bestaand uit één of meerdere elastomeric blokken en één of meerdere thermoplastische blokken en (b) een therapeutische agent, waarin het blokcopolymeer met de therapeutische agent wordt geladen. Het blokcopolymeer is bij voorkeur van formule X -- (AB).sub.n, waar A een elastomeric blok is, B is een thermoplastisch blok, is n een positief geheel aantal en X is een zaadmolecule. De elastomeric blokken zijn bij voorkeur polyolefin blokken, en de thermoplastische blokken worden bij voorkeur geselecteerd uit vinyl aromatische blokken en methacrylate blokken. Volgens een ander aspect van de uitvinding, wordt een medisch hulpmiddel verstrekt, minstens een gedeelte waarvan in het lichaam van een patiënt inlasbaar of inplanteerbaar is. Het medische hulpmiddel bestaat (a) uit het bovengenoemde biocompatibele blokcopolymeer en (b) uit een therapeutische agent, waarin het blokcopolymeer met de therapeutische agent wordt geladen. Volgens een ander aspect van de onderhavige uitvinding, wordt een methode van behandeling verstrekt waarin het bovengenoemde apparaat wordt geïnplanteerd of in een patiënt opgenomen, resulterend in de versie van therapeutische agent in de patiënt over een uitgebreide periode. Volgens nog een ander aspect van de uitvinding, wordt een met een laag bedekt medisch hulpmiddel verstrekt dat bestaat uit: (a) een intravascular of intervascular medisch hulpmiddel en (b) een deklaag meer dan minstens een gedeelte intravascular of intervascular een medisch hulpmiddel, waarin de deklaag uit het bovengenoemde biocompatibele blokcopolymeer bestaat.
