This invention provides a novel class of vitamin D related compounds, namely, the 2-alkylidene-19-nor-vitamin D derivatives, as well as a general method for their chemical synthesis. The compounds have the formula: ##STR1## where Y.sub.1 and Y.sub.2, which may be the same or different, are each selected from the group consisting of hydrogen and a hydroxy-protecting group, R.sub.6 and R.sub.8, which may be the same or different, are each selected from hydrogen, alkyl, hydroxyalkyl and fluoroallly, or when taken together represent the group --(CH.sub.2).sub.X -- where x is an integer from 2 to 5, and where the group R represents any of the typical side chains known for vitamin D type compounds. These 2-substituted compounds are characterized by relatively high intestinal calcium transport activity and relatively high bone calcium mobilization activity resulting in novel therapeutic agents for the treatment of diseases where bone formation is desired, particularly low bone turnover osteoporosis. These compounds also exhibit pronounced activity in arresting the proliferation of undifferentiated cells and inducing their differentiation to the monocyte thus evidencing use as anti-cancer agents and for the treatment of diseases such as psoriasis.

Deze uitvinding verstrekt een nieuwe klasse van vitamine D samenstellingen, namelijk, de àlkylidene19norvitamin D derivaten, evenals een algemene methode voor hun chemische synthese met elkaar in verband bracht. De samenstellingen hebben de formule: ## STR1 ## waar Y.sub.1 en Y.sub.2, die zelfde of verschillend kunnen zijn, elk geselecteerd uit de groep die uit waterstof bestaat zijn en een hydroxy-beschermt groep, een R.sub.6 en een R.sub.8, die zelfde of verschillend kunnen zijn, zijn geselecteerd uit waterstof, alkyl, hydroxyalkyl elk en fluoroallly, of wanneer samen genomen vertegenwoordigen de groep -- (CH.sub.2).sub.X -- waar x een geheel van 2 tot 5 is, en waar de groep R om het even welk van de typische zijketens vertegenwoordigt die voor het type van vitamineD samenstellingen worden gekend. Deze 2-gesubstitueerde samenstellingen worden gekenmerkt door de vrij hoge intestinale activiteit van het calciumvervoer en vrij hoge de mobiliseringsactiviteit van het beencalcium resulterend in nieuwe therapeutische agenten voor de behandeling van ziekten waar de beenvorming, in het bijzonder de lage osteoporose van de beenomzet wordt gewenst. Deze samenstellingen stellen ook uitgesproken activiteit in het arresteren van de proliferatie van niet gedifferentieerde cellen en het veroorzaken van hun differentiatie die aan monocyte tentoon zo van gebruik blijk geeft als agenten tegen kanker en voor de behandeling van ziekten zoals psoriasis.
